Human Adiponectin (Acrp30) is secreted by 3T3-L1 cells.Adiponectin is a secreted protein of 244 amino acids that belongs to a proteinfamily possessing a collagen-like domain through which they form homotrimers,which further combine to make oligomeric complexes. It specificallybinds to collagen type 1, collagen type 3 and collagen type 5. Levels ofAdiponectin are decreased in obesity and it has been observed thatAdiponectin levels are also reduced in patients with coronary artery disease.
The Hoelzel Human Adiponectin ELISA (Enzyme-LinkedImmunosorbent Assay) kit is an in vitro enzyme-linked immunosorbentassay for the quantitative measurement of human Adiponectin in serum,plasma, cell culture supernatants and urine. This assay employs an antibodyspecific for human Adiponectin coated on a 96-well plate. Standards andsamples are pipetted into the wells and Adiponectin present in a sample isbound to the wells by the immobilized antibody. The wells are washed andbiotinylated anti-human Adiponectin antibody is added. After washing awayunbound biotinylated antibody, HRP-conjugated streptavidin is pipetted tothe wells. The wells are again washed, a TMB substrate solution is added tothe wells and color develops in proportion to the amount of Adiponectinbound. The Stop Solution changes the color from blue to yellow, and theintensity of the color is measured at 450 nm.
