Application Notes |
Western blot analysis (0.5-4 ug/ml). However, the optimal conditions should be determined individually. The immunoaffinity-purified antibody detects a >350 kDa and a 180 kDa protein (believed to be a processed form of the catalytic subunit), corresponding to the expected molecular weight of DNA-PKcs, in samples from human, mouse, rat and hamster origins. |
Concentration |
0.5 mg/ml |
Cross-Reactivity |

Description |
Rabbit Anti-Human PRKDC Affinity Purified polyclonal antibody. DNA-dependent protein kinase (DNA-PK) is made up of 3 subunits, 2 Ku proteins and a large catalytic subunit (DNA-PKcs). DNA-PKcs falls into the phosphatidyl-inositol 3-kinase superfamily although it does have serine/threonine protein kinase activity. This subunit migrates on SDS-PAGE gels as an intact and smaller processed form. in vitro , DNA-PK phosphorylates several transcription factors and other DNA-binding proteins and is believed to play a role in DNA damage recognition, repair and transcription. There is evidence to suggest that DNA-PKcs may be a critical target for proteolysis by a caspase in apoptosis. |
Formulation |
100 ug (0.5 mg/ml) immunoaffinity purified rabbit anti-DNA-PK polyclonal antibody in phosphate buffered saline (PBS), pH 7.2, containing 30% glycerol + 0.5% BSA + 0.01% thiiomersal. Precaution: Thiomersal is a poisonous and hazardous substance which should be handled by trained staff only. |
Immunogen |
Synthetic peptide corresponding to residues surrounding amino acid 4088 of human DNA-PK. |
Purification |
Affinity purified |
Storage and Stability |
Store at -20C or in working aliquots at -80C for long term storage. Avoid repeated freeze-thaw cycles. |
Specificity |
Recognizes a > 350 kDa and a 180 kDa protein (believed to be a processed form of the catalytic subunit), corresponding to the expected molecular weight of DNA-PKcs, in samples from human, mouse, rat and hamster origins. |
