Synthetic branched peptide containing the sequence ARme2KSA… in which me2K corresponds to dimethyl-lysine at residue 27 of human histone H3 (KLH).

Histone H3 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure. Acetylation of histone H3 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).

Applications:
Suitable for use in Western Blot, Peptide Inhibition, Immunocytochemistry, Chromatin Immunoprecipitation, Beadlyte Histone-Peptide Specificity Assay and Dot Blot. Other applications not tested.

Recommended Dilution:
Western Blot: 0.5ug/ml detects dimethylated histone H3 in acid extracted proteins from HeLa cells. No detection of 1ug unmethylated recombinant Histone H3 (H5110-12K) was observed.
Peptide Inhibition: 1uM of the immunizing peptide to completely abolishes detection of histone H3 in Western Blot analysis of HeLa acid extracts. Slight signal reduction was observed with peptide containing dimethyl-lysine 9. Signal was not reduced by branched monomethyl H3(Lys27), linear monomethyl H3(Lys27), linear trimethylH3(Lys27), or linear dimethyl H3(Lys4) peptides.
Beadlyte Histone-Peptide Specificity Assay: 12.5ng/ml or a range of dilutions detected peptides containing the dimethyl-lysine modification at residue 27. Peptides containing other modifications were not detected.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. 

Note: Applications are based on unconjugated antibody.
