Description of target
NFKBIA, also called IKBA or MAD-3, is one member of a family of cellular proteins that function to inhibit the NF-ICB transcription factor. It is mapped to 14q13.2. NFKBIA inhibits NF-ICB by masking the nuclear localization signals (NLS) of NF-ICB proteins and keeping them sequestered in an inactive state in the cytoplasm. It moves between the cytoplasm and the nucleus via a nuclear localization signal and CRM1-mediated nuclear export. The effect of the nonpathogenic bacteria is specific to the SCF complex substrates CTNNB1 and NFKBIA. This may help to explain the beneficial effects of treatment of inflammatory bowel disease with nonpathogenic probiotic enteric organisms. In addition, NFKBIA blocks the ability of NF-ICB transcription factors to bind to DNA, which is required for NF-ICB's proper functioning.

Dilution
Ubiquitin-like modifier involved in formation of autophagosomal vacuoles (autophagosomes). Plays a role in mitophagy which contributes to regulate mitochondrial quantity and quality by eliminating the mitochondria to a basal level to fulfill cellular energy requirements and preventing excess ROS production. Whereas LC3s are involved in elongation of the phagophore membrane, the GABARAP/GATE-16 subfamily is essential for a later stage in autophagosome maturation. Promotes primary ciliogenesis by removing OFD1 from centriolar satellites via the autophagic pathway.
