Immunogen:
Synthetic peptide corresponding to the amino acids 1 - 24 of the human ACTH conjugated to Keyhole Limpet Haemocyanin.
Specificity:
Recognises 1-24 ACTH (Synacthen) which as a synthetic analogue of naturally-occurring Adrenocorticotropic Hormone can be used to measure adrenal reserve and for the diagnosis of adrenal insufficiency by acute adrenocortical stimulation. ACTH released from the anterior pituitary gland in response to corticotropin-releasing hormone from the hypothalamus acts on the adrenal cortex to stimulate the production of corticosteroids such as cortisol involved in the response to stress. Studies have shown that the administration of 1-24 ACTH increases blood pressure believed to be attributed to an increase in ACTH-stimulated cortisol secretion in association with increased cardiac output. 0178-0149 does not recognise 17-39 ACTH (CLIP). 0178-0149 cross-reacts with rat N-terminal ACTH. Recommended Secondary Antibodies: Rabbit Anti Mouse IgGGoat Anti Mouse IgGGoat Anti Mouse IgG (H/L)Goat Anti Mouse IgG IgA IgMHuCAL Anti Mouse IgG1Goat Anti Mouse IgG (Fc)Sheep Anti Mouse IgG (H/L)
Function:
ACTH stimulates the adrenal glands to release cortisol. MSH (melanocyte-stimulating hormone) increases the pigmentation of skin by increasing melanin production in melanocytes. Beta-endorphin and Met-enkephalin are endogenous opiates. Subcellular locationSecreted. Tissue specificityACTH and MSH are produced by the pituitary gland. Post-translational modificationSpecific enzymatic cleavages at paired basic residues yield the different active peptides. O-glycosylated; reducing sugar is probably N-acetylgalactosamine. Involvement in diseaseDefects in POMC may be associated with susceptibility to obesity [MIM:601665]. Defects in POMC are the cause of pro-opiomelanocortinin deficiency [MIM:609734]. Affected individuals present early-onset obesity adrenal insufficiency and red hair. Ref. 22Sequence similaritiesBelongs to the POMC family. 1. Willment et al.
