Specificity:
Recognises human prolyl hydroxylase 1 (PHD1) a 44kDa enzyme expressed abundantly in all tissues with the highest expression in testis. Hypoxia inducible factor-1 (HIF-1) is a transcriptional complex consisting of an alpha and beta subunit which plays a key role in coordinating the cellular response to hypoxia. During normal oxygen conditions the alpha subunit of HIF-1 is rapidly degraded however when hypoxia occurs this degradation is suppressed and HIF-1 activates the transcription of various genes important for survival and adaptation to hypoxia. Prolyl hydroxylase 1 catalyses the hydroxylation of specific prolyl residues within the HIF-1 alpha subunit thereby targeting this subunit for degradation. Prolyl hydroxylase 1 might also play a role in the regulation of cell growth. Function:
Catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates HIF-1 alpha at \' Pro-402\' and \' Pro-564\' and HIF-2 alpha. Functions as a cellular oxygen sensor and under normoxic conditions targets HIF through the hydroxylation for proteasomal degradation via the von Hippel-Lindau ubiquitination complex. May play a role in cell growth regulation. Isoform p40 and isoform p43 exhibit the same level of activity. Ref. 9Ref. 12Catalytic activityAn HIF alpha chain L-proline + 2-oxoglutarate + O2 = An HIF alpha chain trans-4-hydroxy-L-proline + succinate + CO2. Cofactor: Binds 1 Fe2+ ion per subunit. Ascorbate. Subunit structureInteracts with the E3 ubiquitin-protein ligase SIAH2 with isoform p40 having a higher propensity to bind than isoform p43; in vitro the interaction targets both isoforms for proteasomal degradation. Ref. 12Subcellular locationCytoplasm. NucleusRef. 10. Tissue specificityExpressed abundantly in all tissues with highest expression in testis. Expressed in hormone responsive tissues including normal and cancerous mammary ovarian and prostate epithelium. Ref. 10Induction: By estrogen. Isoform p43 induced and its protein stability enhanced by hypoxia. Isoform p40 repressed by hypoxia. Isoform p40 and isoform p43 induced by proteasomal inhibitor MG132 (at protein level). Ref. 12Ref. 2Sequence similaritiesContains 1 Fe2OG dioxygenase domain. Sequence cautionThe sequence AAH01723. 1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally shortened.
