Specificity:
CD247NCBI
Gene ID:
919
Specificity:
CD247
Clone:
G3
Immunogen:
113 amino acid synthetic peptide corresponding to the cytoplasmic domain of murine zeta TCR subunit.
Fusion Partner:
Spleen cells from immunised BALB/c mice were fused with cells of the Ag8. 653 mouse myeloma cell line.
Specificity Note:
The T cell antigen receptor (TCR) consists of a ligand-specific alpha/beta heterodimer non-covalently associated with five invariant chains including the CD3 gamma/delta/eta and zeta subunits all of which are required for efficient surface expression. T cell activation through the TCR induces cellular differentiation and/or proliferation and the production of lymphokines and cytokines. Both the CD3 and TCR zeta subunits are proposed to be responsible for the intracellular signal transduction events. A novel monoclonal antibody named G3 has been developed that is specific for the TCR zeta chain subunit (CD247). The antibody is capable of precipitating both non-phosphorylated and tyrosine phosphorylated TCR zeta chain.
Preparation:
Purified IgG prepared by affinity chromatography on Protein A from tissue culture supernatant
Buffer Solution:
Phosphate buffered saline pH7. 2
Preservative Stabilisers:
0. 09%Sodium AzideSuggested
Flow Cytometry:
Membrane permeabilisation is required for this application using a digitonin based method. Suggested
Dilution:
Flow Cytometry - 1/50 - 1/100Immunohistology - Resin - 1ug/mlImmunoprecipitation - 2ug/ml
Function:
Probable role in assembly and expression of the TCR complex as well as signal transduction upon antigen triggering.
Subunit:
The TCR/CD3 complex of T-lymphocytes consists of either a TCR alpha/beta or TCR gamma/delta heterodimer coexpressed at the cell surface with the invariant subunits of CD3 labeled gamma delta epsilon zeta and eta. CD3-zeta forms either homodimers or heterodimers with CD3-eta. Interacts with SLA and SLA2. Interacts with DOCK2 and TRAT1. Interacts with HIV-1 Nef protein. Interacts with SHB.
Subcellular Location:
Membrane; Single-pass type I membrane protein.
Domain:
The ITAM domains mediate interaction with SHB.
Ptm:
Phosphorylated on Tyr residues after T-cell receptor triggering (By similarity).
Disease:
Defects in CD247 are a cause of primary T-cell immunodeficiency [MIM:610163]. Affected individuals suffer of recurrent infections. Patients T-cell counts are very low and B-cell counts are normal.
Similarity:
Belongs to the CD3Z/FCER1G family.
Similarity:
Contains 3 ITAM domains. 1. van Oers. N. S. C. et al. (1994) Production and characterisation of monoclonal antibodies specific for the murine T cell receptor zeta chain. 2. Anderson. P. (1995) mAb reative with lymphocyte restricted intracellular antigens. Leucocyte Typing V p325. Oxford University Press.
