Human recombinant full length cyclin D1 protein. Cellular Localization: Nuclear

Recognizes a protein of 36kD, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.

Applications:
Suitable for use in Immunofluorescence, Flow Cytometry (Not Tested), FLISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.

Recommended Dilution:
Flow Cytometry (Not Tested): 0.5-1ug/million cells
Immunofluorescence: 1-2ug/ml
Western Blot: 0.5-1ug/ml
Immunoprecipitation: 1-2ug/500ug protein lysate
Immunohistochemistry (Frozen & Formalin-fixed): 0.5-1ug/ml for 30 minutes at RT
(Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes)
Optimal dilution for a specific application should be determined.

Positive Control:
T47D, ZR75, BT474, SKBR3, MCF-7, MDA-MB-453, HT29, Ramos, Jurkat or A431 cells. ~60% of mantle cell lymphomas and ~40% of breast carcinomas are positive.

Storage and Stability:
Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer.
Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.

Note: Applications are based on unconjugated antibody.
