Description of target	Non-heme iron-containing dioxygenase that catalyzes the stereo-specific peroxidation of free and esterified polyunsaturated fatty acids generating a spectrum of bioactive lipid mediators. Mainly converts arachidonic acid to (12S)-hydroperoxyeicosatetraenoic acid/(12S)-HPETE but can also metabolize linoleic acid. Has a dual activity since it also converts leukotriene A4/LTA4 into both the bioactive lipoxin A4/LXA4 and lipoxin B4/LXB4. Through the production of specific bioactive lipids like (12S)-HPETE it regulates different biological processes including platelet activation. It also probably positively regulates angiogenesis through regulation of the expression of the vascular endothelial growth factor. Plays a role in apoptotic process, promoting the survival of vascular smooth muscle cells for instance. May also play a role in the control of cell migration and proliferation.

Kit principle	The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Arachidonate-12-Lipoxygenase (ALOX12). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Arachidonate-12-Lipoxygenase (ALOX12). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Arachidonate-12-Lipoxygenase (ALOX12), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm +/- 10nm. The concentration of Arachidonate-12-Lipoxygenase (ALOX12) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Kit reproducibility	Intra-assay Precision (Precision within an assay): 3 samples with low, middle and high level Arachidonate-12-Lipoxygenase (ALOX12) were tested 20 times on one plate, respectively. Inter-assay Precision (Precision between assays): 3 samples with low, middle and high level Arachidonate-12-Lipoxygenase (ALOX12) were tested on 3 different plates, 8 replicates in each plate.
