It needs to form a complex with a corepressor protein (Groucho) to repress transcription. Transcription factor HES-1 has a particular type of basic domain which contains a helix-interrupting proline, that binds to the N-box (CACNAG) rather than the canonical E-box (CANNTG). It also consists of a C-terminal WRPW motif that is necessary for the interaction with Groucho. It localises to the nucleus.
