Application notes
Specific for endogenous levels of the ~78 kDa synapsin I doublet phosphorylated at Ser62, 67. Immunolabeling is blocked by preadsorption with the phosphopeptide used as antigen, but not by the corresponding non-phosphopeptide.

Gene Full Name
Synapsin-1
Background
Synapsin I plays a key role in synaptic plasticity in brain (Feng et al., 2002; Nayak et al., 1996). This effect is due in large part to the ability of the synapsins to regulate the availability of synaptic vesicles for release. The role of synapsin in synaptic plasticity and in synaptogenesis is regulated by phosphorylation (Jovanovic et al., 2001; Kao et al., 2002). Ser-549 along with Ser-62 and Ser-67 are the sites of Synapsin I that are phosphorylated by MAP kinase (Czernik et al., 1987; Jovanovic et al., 1996).
Immunogen
Synthetic phospho-peptide corresponding to amino acid residues surrounding Ser62/67 of rat synapsin, conjugated to keyhole limpet hemocyanin (KLH). Immunogen species is Rat.
Contents
10 mM HEPES (pH 7.5), 150 mM NaCl, 100 µg per ml BSA and 50% glycerol.
Purification
Prepared from pooled rabbit serum by affinity purification via sequential chromatography on phospho and non-phosphopeptide affinity columns.
Concentration
0.5-1mg/ml, actual concentration vary by lot. Use suggested dilution ratio to decide dilution procedure.
Phospho Site
Phosphorylated, Ser62, 67

Protein Name
Synapsin-1
Protein Function
Neuronal phosphoprotein that coats synaptic vesicles, binds to the cytoskeleton, and is believed to function in the regulation of neurotransmitter release. Regulation of neurotransmitter release. The complex formed with NOS1 and CAPON proteins is necessary for specific nitric-oxide functions at a presynaptic level.
Subcellular Localization
Cell junction, synapse. Golgi apparatus.
