CD289 (N-TERMINAL)
Isotype:
Polyclonal IgGSpecies Cross Reactivity: Reacts with: MouseN. B. Antibody reactivity and working conditions may vary between species.
Buffer Solution:
Phosphate buffered saline
Preservative Stabilisers:
0. 09% Sodium Azide (NaN3) 0. 1% Bovine Serum AlbuminApprox. Protein Concentrations: IgG concentration 2. 0mg/ml
Immunogen:
Synthetic peptide SLKYNNLTVVPRNLPSSLEYLLLSYNRIVKL corresponding to amino acids 205-235 within the N-terminal domain of human CD289.
Specificity:
Specifically recognises an epitope within the N-terminal (NT) region of CD289 otherwise known as Toll-like receptor 9 (TLR9) a type I transmembrane glycoprotein and member of the toll-like receptor family which triggers MyD88 and TRAF6 resulting in NF-kappaB activation cytokine secretion and the inflammatory response. CD289 acts as a receptor for unmethylated cytidine-phosphate-guanosine dinucleotides in bacterial DNA (CpG-DNA) playing a primary role in pathogen recognition and activation of innate immunity.
Specificity:
Is reported as suitable for use in immunocytochemistry on acetone fixed cells. Recommended Secondary Antibodies: Rabbit Anti Goat IgG (Fc)
Function:
Key component of innate and adaptive immunity. TLRs (Toll-like receptors) control host immune response against pathogens through recognition of molecular patterns specific of microorganisms. TLR9 is a nucleotide-sensing TLR which is activated by unmethylated cytidine-phosphate-guanosine (CpG) dinucleotides. Acts via MYD88 and TRAF6 leading to NF-kappa-B activation cytokine secretion and the inflammatory response By similarity. Ref. 9Subunit structureInteracts with MYD88 via their respective TIR domains By similarity. Interacts with CNPY3 By similarity. Interacts (via transmembrane domain) with UNC93B1 By similarity. Subcellular locationEndoplasmic reticulum membrane; Single-pass type I membrane protein By similarity. Endosome By similarity. Lysosome By similarity. Cytoplasmic vesicle & rsaquo; phagosome By similarity.
Note:
Relocalizes from endoplasmic reticulum to endosome and lysosome upon stimulation with agonist By similarity. Tissue specificityHighly expressed in spleen lymph node tonsil and peripheral blood leukocytes especially in plasmacytoid pre-dendritic cells. Levels are much lower in monocytes and CD11c+ immature dendritic cells. Also detected in lung and liver. Sequence similaritiesBelongs to the Toll-like receptor family. Contains 26 LRR (leucine-rich) repeats. Contains 1 TIR domain. 1. Letarte M. et al. (1986) Correlations between the 44D7 antigenic complex and the plasma membrane Na+-Ca2+ exchanger. Biochem. Cell.
