Genway ID:
GWB-3F33C1
Isotype:
IgG
Immunogen:
Synthetic peptide (Human) - which represents a portion of human Replication Factor C 4 37 kDa subunit encoded within exon 2.
Antigen Species:
Human
Positive Control:
Nuclear extract from HeLa cells
Target:
RFC4
Concentration:
1 mg/ml Storage
Buffer:
Tris-citrate/phosphate pH 7-8 0. 1% sodium azide
Application Note:
IP: Use at a concentration of 2 - 4 µ g/ml. WB: Use at a dilution of 1/1000 - 1/10000. Predicted
Molecular Weight:
37 kDa. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. The elongation of primed DNA templates by DNA polymerase delta and DNA polymerase epsilon requires the action of 2 accessory proteins proliferating cell nuclear antigen (PCNA) and Activator 1 (A1; also called Replication Factor C). A1 is an enzyme that contains 5 different subunits of 140 40 38 37 and 36 kD. Chen et al. (1992) isolated the gene encoding the 37-kD subunit from HeLa cells. The deduced amino acid sequence showed a high degree of homology to the 40-kD subunit of A1 but unlike the 40-kD protein the 37-kD expressed protein did not bind ATP. Other findings suggested that both the 37- and 40-kD subunits of A1 are required for the biologic role of A1 and that they may function differently in this process.
Function:
The elongation of primed DNA templates by DNA polymerase delta and epsilon requires the action of the accessory proteins proliferating cell nuclear antigen (PCNA) and activator 1. This subunit may be involved in the elongation of the multiprimed DNA template.
Subunit:
Heterotetramer of subunits RFC2 RFC3 RFC4 and RFC5 that can form a complex either with RFC1 or with RAD17. The former interacts with PCNA in the presence of ATP while the latter has ATPase activity but is not stimulated by PCNA.
Subcellular Location:
Nucleus (Probable).
Miscellaneous:
Despite of the presence of a putative ATP-binding motif this protein does not bind ATP.
Similarity:
Belongs to the activator 1 small subunits family.
