Immunogen:
A peptide corresponding to amino acids near the middle of human MD-2.
Specificity:
Recognises MD-2 a member of the Toll/interleukin-1 receptor (TIR) family a group of proteins which include Toll like receptors. TLR4 requires the binding of MD-2 to its extracellular region for best response to bacterial LPS. MD-2 also enhances TLR4-dependent activation of NF-kappa-beta and cooperates with TLR2 in the response to cell wall components from bacteria. Function:
Cooperates with TLR4 in the innate immune response to bacterial lipopolysaccharide (LPS) and with TLR2 in the response to cell wall components from Gram-positive and Gram-negative bacteria. Enhances TLR4-dependent activation of NF-kappa-B. Cells expressing both MD2 and TLR4 but not TLR4 alone respond to LPS. Subunit structureHeterogeneous homopolymer formed from homodimers; disulfide-linked. Belongs to the lipopolysaccharide (LPS) receptor a multi-protein complex containing at least CD14 LY96 and TLR4. Binds to the extracellular domains of TLR2 and TLR4. Ligand binding induces interaction with TLR4 and oligomerization of the complex.
