Terminal deoxynucleotidyltransferase (TDT) is a DNA polymerase that enhances Ig and TcR gene diversity in the N region at the junctions of variable diversity and joining segments in B- and T-cells. TDT synthesizes the N region in concert with many proteins including DNA-PKcs Ku70 and Ku86. It is expressed in immature lymphocytes of the thymus and bone marrow (1-3). Because TDT contributes to the generation of junctional diversity in antigen receptors of immature lymphocytes its expression serves as a developmental marker for the sites of teleost lymphopoiesis (4). TDT forms a ternary complex with TdIF2 and core histone where the enzymatic consequence of this complex is the reduction of TDT activity (2). TDT is an oligoclonal marker characterizing several populations of undifferentiated or poorly differentiated blasts that tend to develop towards or along the lymphoid pathway. Together with specific immunological markers this enzyme is useful to define the particular type of leukemic cells (5). 1.
