Application Note:
IHC-P: Use at a concentration of 1 - 2 µ g/ml. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer pH 6. 0 for 10-20 min followed by cooling at room temp for 20 minIP: Use at a concentration of 2 µ g/ml. Denatured verified. Use Protein G. WB: Use at a concentration of 1 µ g/ml for 2 hours at room temp. Detects a band of approximately 34 kDa. Not tested in other applications. Localization:
Cytoplasmic Caspases play a major role in the transduction of the apoptotic signal and execution of apoptosis in mammalian cells. Caspase 3 (CPP32) and Caspase 6 (Mch2) are the major active caspases in apoptotic cells and are activated in response to distinct apoptosis inducing stimuli and in all cell lines analyzed. Both CPP32 and Mch2 are present in apoptotic cells as multiple active species. Caspase 6 cleaves nuclear mitotic apparatus protein (NuMA) and mediates the shrinkage and fragmentation of nuclei.
Function:
Involved in the activation cascade of caspases responsible for apoptosis execution. Cleaves poly(ADP-ribose) polymerase in vitro as well as lamins. Overexpression promotes programmed cell death.
Catalytic Activity:
Strict requirement for Asp at position P1 and has a preferred cleavage sequence of Val-Glu-His-Asp-|-.
Enzyme Regulation:
Activation is suppressed by phosphorylation at Ser-257.
Subunit:
Heterotetramer that consists of two anti-parallel arranged heterodimers each one formed by a 18 kDa (p18) and a 11 kDa (p11) subunit.

Cytoplasm.
Ptm:
Cleavages by caspase-3 caspase-8 or -10 generate the two active subunits.
