Vascular endothelial growth factor receptor-1 (VEGF R1), also known as Fms-like tyrosine kinase 1 (FLT-1), tyrosine-protein kinase FRT, tyrosine-protein kinase receptor FLT (FLT), vascular permeability factor receptor, is a receptor tyrosine kinase (RTK) specific for the angiogenic factors VEGF (VEGF-A), VEGF-B and PlGF. Two major isoforms of human VEGF R1 have been identified. The full length VEGF R1 is a glycoprotein composed of seven extracellular immunoglobulin (Ig)-like domains, a membrane-spanning region, and an intracellular tyrosine kinase domain with a kinase insert sequence. The truncated, soluble form of VEGF R1(sVEGF R1) consists of only the first six extracellular Ig-like domains in which the first three N-terminal Ig-like domains are ligand binding sites and the fourth Ig-like domain is responsible for receptor dimerization, which is a prerequisite for activation through trans-phosphorylation. VEGF R1 can form homodimers as well as active heterodimers with VEGF R2, while the sVEGF R1 forms inactive heterodimers with VEGF R2. VEGF mediated activation of VEGF R1 and VEGF R2 is essential to both normal and pathological angiogenesis. VEGF R1 is highly expressed on monocyte/macrophage lineages, whereas expression of both VEGF receptors is primarily restricted to endothelial cells and is induced by various stimuli such as VEGF, FGF basic, PECAM-1, cell-cell contact, and hypoxia.
