Glycogen Synthase Kinase 3 (GSK-3) is a member of the Ser/Thr kinase family. GSK-3 functions as a negative regulator in the hormonal control of glucose homeostasis, Wnt signaling and regulation of transcription factors and microtubules, by phosphorylating and inactivating glycogen synthase (GYS1 or GYS2), CTNNB1/beta-catenin, APC and AXIN1. There are two mammalian GSK-3 isoforms encoded by distinct genes, GSK-3 alpha and GSK-3 beta, which are structurally similar, but functionally differently. GSK-3a is inhibited by phosphorylation at S21 by Akt and other kinases. GSK-3 alpha and GSK-3 beta share 85% amino acid identity. GSK3B is involved in energy metabolism, neuronal cell development, and body pattern formation. Dysregulated GSK-3 has been implicated in several diseases including type II diabetes, Alzheimers disease, bipolar disorder, and cancer.
