The cAMP ELISA kit applies a competitive enzyme immunoassay technique. The tested sample, cAMP standard and Alkakine phosphatase (AP)-conjugated cAMP together with cAMP specific rabbit IgG are added into the microplate wells pre?coated with the anti?rabbit IgG antibody and incubated appropriately. The wells are then decanted and washed with wash buffer. The wells are further incubated with a substrate (pNpp) for AP and a stop solution is added to terminate the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 405 nm in a microplate reader. The intensity of the color is inversely proportional to the cAMP concentration, as cAMP from sample and AP cAMP conjugate compete for the anti?cAMP antibody binding site.
