Integrin alpha2beta1 is one of twelve integrin family adhesion receptors that share the beta1 subunit (1­3). It is the non­covalent heterodimer of 160kD alpha2 (CD49b) and 130kD beta1 (CD29) type I transmembrane glycoprotein subunits. It is one of six very late antigens on activated T cells, designated VLA2 (3). The alpha2 extracellular domain (ECD) contains an I (inserted) domain which includes the ligand binding site (2, 3). The beta1 ECD contains a vWFA domain, which participates in binding. Each subunit then has a transmembrane sequence and a short cytoplasmic tail. The dimer is folded when it is least active. Divalent cations and intracellular (inside­out) signaling convert it to its most active, extended and open conformation (1, 2). The 1103 amino acid (aa) mouse alpha2 extracellular domain (ECD) shares 93% aa sequence identity with rat and 80­81% with human, canine, bovine and equine alpha2, while the 708 aa mouse beta1 ECD shares 98% aa identity with rat and 93­94% with human, bovine, porcine, ovine, canine and feline beta1. The I domain­containing beta1 integrins (alpha1beta1, alpha2beta1, alpha10beta1 and alpha11beta1) all bind collagens, with alpha2beta1 preferring collagens I­III (4, 5). Platelet alpha2beta1, also called GPIa, cooperates with another adhesion protein, GPVI, to coordinate platelet collagen binding and activation (3, 6, 7). Other alpha2beta1 ligands include laminin, decorin, E­cadherin, and collagen­like regions of collectin molecules such as C1q (4). Adhesion is synergized by crosstalk with syndecan­1 or HGF R/c­Met, and antagonized by crosstalk with integrin alpha1beta1 (8­10). In addition to expression on selected hematopoietic cells, alpha2beta1 is present on a wide variety of non­hematopoietic cells (4). Mice deficient in the alpha2 subunit have defects in innate immune responses, wound mast cell infiltration and angiogenesis, and platelet responses to collagen (6, 11, 12). In innate immunity, alpha2beta1 binding to C1q initiates the complement cascade and co­stimulates mast cell activation, triggering neutrophil influx (4, 12).

Activity:
Measured by its binding ability in a functional ELISA. When bovine collagen-II is coated at 10ug/mL, Recombinant Mouse Integrin alpha 2 beta 1 binds with an apparent KD <1nM.

Storage and Stability:
Lyophilized and reconstituted products are stable for 6 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
