DNA topoisomerases I and II are a class of nucleases, with type II including two highly homologous isozymes: topoisomerase IIα and IIβ.These enzymes are capable of regulating the topological structure of DNA, maintaining genome integrity, and playing important roles in DNAreplication, recombination, transcription, and chromosome segregation. Topoisomerase I can cleave and reconnect one strand of doublestranded DNA, while topoisomerase II can temporarily break the double-stranded DNA and reconnect the broken ends. Topoisomerases arehighly sensitive to various stresses. Under acidic pH or oxidative stress, topoisomerases can convert into DNA cleavage nucleases, leadingto genomic instability and cell death. Certain topoisomerase-targeting drugs for DNA damage, such as etoposide, can also converttopoisomerases into nucleases. These enzymes are typically trapped as an intermediate product, capable of covalently binding to the 5' endof the cleaved DNA chain. Research has shown that this intermediate product can cause genomic instability and cell death. Therefore, targetingtopoisomerases has become an important direction in the development of cancer chemotherapy drugs.
