Supplied as a liquid in 50mM sodium phosphate, pH 7.5, 100mM potassium chloride, 150mM sodium chloride, 0.5 mg/ml Gentamicin Sulfate.

YT-1 acute human leukemia cells.

Human CD122 (IL-2 R beta) associates with CD25 (IL-2R alpha) to form a high affinity receptor for IL-2 and is expressed on activated T cells, B cells and monocytes. The cytoplasmic domain of CD122 is involved with signal transduction.

Applications:
Suitable for use in Flow Cytometry. Other applications not tested.

Recommended Dilution:
Flow Cytometry: 20ug/ml. 80ul labels 5x10e5 ficoll prepared human peripheral blood lymphocytes.
Optimal dilutions to be determined by the researcher.

Flow Cytometry Protocol:
1. Five x 10e5 ficoll prepared human peripheral blood lymphocytes were stimulated by incubating 5 days with 5ug/ml PHA-P (P3370-61).
2. Stimulated cells were harvested and incubated 45 minutes on ice with 80ul of C2460-01F at 20ug/ml.
3. Cells were washed twice and incubated with I1904-04E: IgG Pab Goat x Mouse (FITC), after which they were washed three times, fixed and analyzed by FACS.
4. Cells stained positive with a mean shift of 0.17 log10 fluorescent units when compared to a Mouse IgG2a negative control at a similar concentration.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
