Recombinant human HSP60 protein. Cellular Localization: Mitochondria in cytoplasm

Recognizes a 60kDa protein, identified as the heat shock protein 60 (hsp60). A wide variety of environmental and pathophysiological stressful conditions trigger the synthesis of a family of proteins known as heat shock proteins (hsp’s), more appropriately called as stress response proteins (srp’s). hsp60 is a potential antigen in a number of autoimmune diseases. In human arthritis and in experimentally induced arthritis in animals, disease development coincides with the development of immune reactivity directed against not only bacterial hsp60, but also against its mammalian homolog. Clone GROEL730 reacts only with human and is useful in distinguishing human hsp60 from other mammalian and bacterial hsp60.

Applications: 
Suitable for use in Immunofluorescence, Flow Cytometry, ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested.

Recommended Dilution:
Flow Cytometry: 0.5-1ug/million cells
Immunofluorescence: 0.5-1ug/ml
Western Blot: 0.25-0.5ug/ml
Immunoprecipitation: 0.5-1ug/500ug protein lysate
Immunohistochemistry: Frozen & Formalin-fixed: 0.5-1ug/ml for 30 minutes at RT
(Staining of formalin-fixed tissues is enhanced by boiling tissue sections in 1mM EDTA Buffer, pH 8.5-9.5, for 10-20 min followed by cooling at RT for 20 minutes
Optimal dilutions to be determined by the researcher.

Positive Control:
HeLa or HepG2 cells. Synovial biopsies from patients with juvenile chronic arthritis. Synovial lining layer is strongly positive for hsp60. Breast carcinoma.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
