Quality Tested by Leinco Western Blotting: To detect Human HCMV IL-10 this biotin conjugate can be used at a concentration of 0.1 - 0.2 µg/ml. This biotin conjugate should be used in conjunction with compatible second-step reagents such as PN:A106 and a chromogenic substrate such as PN:T343. The detection limit for Human HCMV IL-10 is 2 ng/lane under either reducing or non-reducing conditions. The sensitivity of detection may increase up to 50 fold when a chemiluminescent substrate is used.

Human cytomegalovirus (HCMV) is a widespread pathogen that establishes lifelong latent infection facilitated by numerous mechanisms for modulating the host immune system. The UL111A region of the HCMV genome encodes a homolog of human cellular IL-10 (hIL-10). The viral cytokine, cmvIL-10, exhibits many of the immunosuppressive properties of hIL-10. CMVIL-10 has the ability to enhance B cell proliferation.1 CMV IL10 is expressed in CMV-infected cells and can bind to the human IL10 receptor and can compete with human IL10 for binding sites. CMV IL10 requires both subunits of the IL10 receptor complex to induce signal transduction events and biological activities. CMV IL10 inhibits the proliferation of mitogen-stimulated peripheral blood lymphocytes with the potency of human IL10. It also inhibits cytokine synthesis by these cells. Human CMV IL10 also reduces the cell-surface expression of MHC class 1 and MHC class 2 proteins on peripheral blood mononuclear cells.2
