Neutralization:
To yield one-half maximal inhibition [ND50] of the biological activity of h-RANTES (100ng/mL), a concentration of 5-7 μ g/mL of this antibody is required.

ELISA:

In a sandwich ELISA (assuming 100 μ L/well), a concentration of 2.0-4.0 μ g/mL of this antibody will detect at least 500 pg/mL of recombinant human RANTES when used with our biotinylated antigen affinity purified anti-human RANTES (XP-5257Bt) as the detection antibody at a concentration of approximately 0.5-1.0 μ g/mL.

Western Blot:

To detect hRANTES by Western Blot analysis this antibody can be used at a concentration of 0.5-1.0 μ g/mL. Used in conjunction with compatible secondary reagents the detection limit for recombinant hRANTES is 0.25-2.0 ng/lane, under reducing conditions and 0.25-0.50 ng/lane, under non-reducing conditions.

This antibody stained formalin-fixed, paraffin-embedded sections of human tonsil. The recommended concentration is 10.0 ug/ml with an overnight incubation at 4˚C. An HRP-labeled polymer detection system was used with a DAB chromogen. Heat induced antigen retrieval with a pH 6.0 citrate buffer is recommended. Optimal concentrations and conditions may vary.
