Immunogen:
Recombinant human hexokinase 1 (amino acids 1-917) purified from Escherichia coli.
Specificity:
Specifically recognises Hexokinase 1 a 102kDa major allosteric enzyme of the glycolytic pathway which catalyses the phosphorylation of glucose to form glucose-6-phosphate (Glc-6-P). Hexokinase 1 is the first of four distinct mammalian hexokinase isozymes (hexokinase type I to IV) which is expressed in the outer membrane of mitochondria and is regulated (inhibited) by its product Glc-6-P. A deficiency in hexokinase 1 is implicated in the rare hereditary autosomal recessive disease known as nonspherocytic haemolytic anaemia. Its hydrophobic N-terminal sequence may be involved in membrane binding. Tissue specificityIsoform 2 is erythrocyte specific. Isoform 3 and isoform 4 are testis-specific.
Domain:
The N- and C-terminal halves of this hexokinase show extensive sequence similarity to each other. The catalytic activity is associated with the C-terminus while regulatory function is associated with the N-terminus. Each domain can bind a single glucose and Gluc-6-P molecule. Hexokinase deficiency is a rare autosomal recessive disease with nonspherocytic hemolytic anemia as the predominant clinical feature.
