Phosphate buffered saline
Preservative Stabilisers:
0. 02% Sodium Azide (NaN3)Approx. Protein Concentrations: IgG concentration 1. 0mg/ml
Immunogen:
A 16 amino acid peptide from human Caspase-7 amino-terminus
Specificity:
Recognises an epitope within the N-terminal region (NT) of Caspase-7 a 35kDa cysteine protease otherwise known as ICE-like Apoptotic Protease 3 (ICE-LAP3). Caspase-7 a member of the ICE/Ced-3 subfamily is an executioner caspase which undergoes proteolytic cleavage of its precursor to form active p12 and p20 subunits. Evidence that activation of Caspase-7 occurs during cell death induced by the cytokine death receptors Fas/APO-1 and the receptor of tumour necrosis factor (TNFR-1) coupled with the fact that granzyme-B activated Caspase-7 cleaves the nuclear enzyme poly (ADP-ribose) polymerase (PARP) suggests an important role for Caspase-7 in both cytokine-mediated and Granzyme-B mediated apoptosis pathways. Recommended Secondary Antibodies: Sheep Anti Rabbit IgGGoat Anti Rabbit IgG (Fc)Goat Anti Rabbit IgG (H/L)
Function:
Involved in the activation cascade of caspases responsible for apoptosis execution. Cleaves and activates sterol regulatory element binding proteins (SREBPs). Proteolytically cleaves poly(ADP-ribose) polymerase (PARP) at a \' 216-Asp-|-Gly-217\' bond. Overexpression promotes programmed cell death. Catalytic activityStrict requirement for an Asp residue at position P1 and has a preferred cleavage sequence of Asp-Glu-Val-Asp-|-. Enzyme regulationInhibited by isatin sulfonamides. Subunit structureHeterotetramer that consists of two anti-parallel arranged heterodimers each one formed by a 20 kDa (p20) and a 11 kDa (p11) subunit. Ref. 13Subcellular locationCytoplasm. Tissue specificityHighly expressed in lung skeletal muscle liver kidney spleen and heart and moderately in testis. No expression in the brain. Post-translational modificationCleavages by granzyme B or caspase-10 generate the two active subunits. Propeptide domains can also be cleaved efficiently by caspase-3. Active heterodimers between the small subunit of caspase-7 and the large subunit of caspase-3 and vice versa also occur. Ref. 10Sequence similaritiesBelongs to the peptidase C14A family. 1.
