The predicted size of prolactin is 25 kDa. The affinity purified recombinant human prolactin showed two bands: the major band is about 25 kDa, and the minor band is about 27 kDa. Both bands were confirmed by Western blotting using anti-prolactin antibody. The 27-kDa band may represent N-glycosylated and phosphorylated form.

Purified proteins were untreated (Lane 2) or treated with protein deglycosylation enzymes under native (Lane 3) or reducing (Lane 4) conditions.
