Genway ID:
GWB-9B152F
Target:
TFAM is a mitochondrial transcription factor that is a key activator of mitochondrial transcription as well as a participant in mitochondrial genome replication. Gene Symbol: Tfam
Antigen Specificity:
Polyclonal antibody produced in rabbits immunized with a synthetic peptide corresponding to a region of Mouse Tfam. Western Blot: Antibody
Dilution:
0. 5 microg/ml IHC: 4-8ug/ml
Handling:
Add 50ul of distilled water to this antibody before use.
Immunogen:
Synthetic peptide sequence derived from: FQEAKDDSAQGKLKLVNEAWKNLSPEEKQAYIQLAKDDRIRYDNEMKSWE
ELISA Titre:
1:312500
Note:
Suggested starting concentrations are provided. Optimal dilutions should be determined by end-user. Differences in calculated versus apparent molecular weight may be due to post-translational modifications or protein hydrophobicity. Studies in mice have demonstrated that this protein is required to regulate the mitochondrial genome copy number and is essential for embryonic development. A mouse model for Kearns Sayre syndrome was produced when expression of the gene was eliminated by targeted disruption in heart and muscle cells.
Function:
The mitochondrial isoform is involved in mitochondrial transcription regulation. Required for accurate and efficient promoter recognition by the mitochondrial RNA polymerase. Activates transcription by binding immediately upstream of transcriptional start sites. Is able to unwind and bend DNA. The testis-specific isoform may also function as a transcriptional activator or may have a structural role in the compaction of nuclear DNA during spermatogenesis.
Subunit:
Interacts with TFB1M and TFB2M (By similarity).
Subcellular Location:
Nucleus. Mitochondrion.
Tissue Specificity:
The mitochondrial isoform is widely expressed while the nuclear isoform is testis-specific.
Similarity:
Contains 2 HMG box DNA-binding domains. Noack. H. . (2006) Biochim. Biophys. Acta 1760 (2). 141-150.
