Application Note:
WB: Use at a dilution of 1/1000. A starting antibody dilution of 1/5000 is recommended for chemiluminescent substrates. Higher antibody dilutions may be necessary for non-human samples. Full length ADAMTS4 (837 amino acids) has a predicted mass of 90kDa but glycosylation and the abundance of cysteine residues gives ADAMTS4 a greater apparent molecular weight under reducing conditions. The antibody detects the zymogen form at 98 kDa activated forms at 64 kDa (major bands) and breakdown products at 30 kDa. A band at approximately 29 kDa is detected in cell culture media which may be the propeptide domain shed after furin cleavage. EDTA/EGTA treatment of tissues or lysates is required to see latent zymogen. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. Cellular
Localization:
Secreted. Associated with the extracellular matrix. A disintegrin like and metalloproteinase thrombospondin type 1 motif 4 (ADAMTS4 or aggrecanase 1) is a member of the ADAMTS family of multidomain proteins that has demonstrated metalloproteinase functions. The ADAMTS4 protein contains a prohormone convertase cleavage site thrombospondin 1 like domain cysteine rich domain and a propeptide domain. ADAMTS4 activity had been shown to be blocked by TIMP1 2 3 and 4.
Function:
Cleaves aggrecan a cartilage proteoglycan and may be involved in its turnover. May play an important role in the destruction of aggrecan in arthritic diseases. Could also be a critical factor in the exacerbation of neurodegeneration in Alzheimer disease. Cleaves aggrecan at the \' 392-Glu-|-Ala-393\' site.
Catalytic Activity:
Glutamyl endopeptidase; bonds cleaved include 370-Thr-Glu-Gly-Glu-|-Ala-Arg-Gly-Ser-377 in the interglobular domain of mammalian aggrecan. Cofactor: Binds 1 zinc ion per subunit (By similarity).

Secreted extracellular space extracellular matrix (By similarity).
Tissue Specificity:
Expressed in brain lung and heart. Expressed at very low level in placenta and skeletal muscles.

By interleukin-1.

The spacer domain and the TSP type-1 domains are important for a tight interaction with the extracellular matrix.
Domain:
The conserved cysteine present in the cysteine-switch motif binds the catalytic zinc ion thus inhibiting the enzyme. The dissociation of the cysteine from the zinc ion upon the activation-peptide release activates the enzyme.
