IHC: Use at a dilution of 1/200 - 1/1000. WB: Use at a dilution of 1/200 - 1/1000. Detects a band of approximately 46. 3 kDa. Preparation InstructionsMethod 1 for immunostaining and immunoblotting (Western blot)1. Carefully resuspend antibody pellet to uniformity. 2. Remove a fixed amount of suspension and dissolve 1/5 in PBS or TBS to yield a 100 µ g/ml solution. Method 2 for immunoprecipitation supershift immunostaining and immunoblotting (Western blot)1 Pellet antibodies at 10 000 - 15 000 x g for 10 minutes at 2 to 8 C using a microcentrifuge. 2 Carefully remove as much supernatant as possible. It is not necessary to remove all the ammonium sulfate solution; a small residual amount will not effect the antibody preparation. Dissolve the pellet (antibody) in small volume (50µ L) of PBS (or TBS) at final concentration of 1 mg/ml (50µ g/50µ l). Do not allow the pellet to dry out. This can cause loss of activity. Gently allow pellet to dissolve at least 1 hour before use. Do not vortex. Mix by finger-tapping or gentle stirring. Not tested in other applications. Localization:
Nuclear DNA double-strand breaks can be generated by exposure to ionizing radiation or through endogenous cellular reactions. Double-strand breaks are generally thought to be repaired via one of two distinct mechanisms: homologous recombination (HR) or non-homologous end joining (NHEJ). Homologous recombination requires the activity of the members of the RAD52 epistasis group which include RAD50 RAD51 RAD52 RAD54 RDH54/TID1 RAD55 RAD57 RAD59 MRE11 and XRS2. RAD52 can catalyze strand annealing reactions and can form oligomers that attach to ends of single-stranded DNA.

Involved in double-stranded break repair. Plays a central role in genetic recombination and DNA repair by promoting the annealing of complementary single-stranded DNA and by stimulation of the RAD51 recombinase.
