Specificity:
BMI-1NCBI
Gene ID:

Specificity:
BMI-1
Immunogen:
15 amino acid peptide sequence near the centre of human BMI-1.
Specificity Note:
This product recognises 37kDa polycomb complex protein BMI-1 involved in segment specification cell growth and maintenance transcriptional regulation and chromatin modification. BMI-1 is a proto-oncogene which cooperates with the myc oncogene to produce B-lymphomas. It is required in the regulation of normal adult and leukaemic stem cells making BMI-1 a potential target for stem cell proliferation and anti-cancer therapies. BMI-1 represses transcription at the ink4 locus which contains tumour suppressors p16 and p19Arf. BMI-1 is a component of the Polycomb group (PcG) multiprotein PRC1 complex which ubiquitinates histone-2A at lys119 resulting in a heritable change in the ability of chromatin to express genes. BMI-1 is also required to stimulate the E3 ubiquitin ligase activity of RNF2/RING2 within the PRC1 complex.
Antiserum Preparation:
Antisera to human BMI-1 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG prepared by affinity chromatography.
Buffer Solution:
Phosphate buffered saline
Preservative Stabilisers:

Dilution:
Western Blotting - 0. 5 - 1. 0ug/ml
Function:
Component of the Polycomb group (PcG) multiprotein PRC1 complex a complex required to maintain the transcriptionally repressive state of many genes including Hox genes throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A \' Lys-119\' rendering chromatin heritably changed in its expressibility. In the PRC1 complex it is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2.
Subunit:
Component of chromatin-associated class II polycomb repressive complex 1 (PRC1/hPRC-H) at least composed of PCGF2/RNF110 BMI1/PCGF4 CBX2/M33 CBX4/PC2 CBX8/PC3 PHC1 PHC2 PHC3 SCMH1 RING1 and RNF2/RING2. Also component of E3 ubiquitin ligase complex or human polycomb repressive complex 1-like at least composed of PHC2 BMI1 RING1 and RNF2. Interacts with SPOP. Part of a complex consisting of BMI1 CUL3 and SPOP.
Subcellular Location:
Nucleus.
Ptm:
May be polyubiquitinated; which does not lead to proteasomal degradation.
Disease:
Cooperates with the MYC oncogene to produce B-lymphomas.
Similarity:
