Specificity Note:
This product recognises 37kDa polycomb complex protein BMI-1 involved in segment specification cell growth and maintenance transcriptional regulation and chromatin modification. BMI-1 is a proto-oncogene which cooperates with the myc oncogene to produce B-lymphomas. It is required in the regulation of normal adult and leukaemic stem cells making BMI-1 a potential target for stem cell proliferation and anti-cancer therapies. BMI-1 represses transcription at the ink4 locus which contains tumour suppressors p16 and p19Arf. BMI-1 is a component of the Polycomb group (PcG) multiprotein PRC1 complex which ubiquitinates histone-2A at lys119 resulting in a heritable change in the ability of chromatin to express genes. BMI-1 is also required to stimulate the E3 ubiquitin ligase activity of RNF2/RING2 within the PRC1 complex.

Function:
Component of the Polycomb group (PcG) multiprotein PRC1 complex a complex required to maintain the transcriptionally repressive state of many genes including Hox genes throughout development. PcG PRC1 complex acts via chromatin remodeling and modification of histones; it mediates monoubiquitination of histone H2A \' Lys-119\' rendering chromatin heritably changed in its expressibility. In the PRC1 complex it is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2.
