Specificity:
This antibody recognizes the human c-kit cell surface antigen. c-Kit is expressed by some haematopoietic stem cells (approx. 70% of CD34+ve cells in normal bone marrow are CD117+ve) by some acute myeloid leukaemias and by mast cells. Functionally c-Kit is a receptor for stem cell factor and has receptor tyrosine kinase activity. This antibody is non-neutralizing.

Application Note:
For FACS: Use at a dilution of 1:2-1:20. Use 10 µ l of the suggested working dilution to label 10^6 cells in 100 µ l. Not tested in other applications. Optimal dilutions/concentrations should be determined by the researcher. c-Kit also known as CD117 and stem cell factor receptor is a 145 kDa transmembrane tyrosine kinase encoded by the c-Kit proto-oncogene. c-Kit acts to regulate a variety of biological responses including cell proliferation apoptosis chemotaxis and adhesion. Ligand binding to the extracellular domain leads to autophosphorylation on several tyrosine residues within the cytoplasmic domain and activation. Mutations in c Kit have been found to be important for tumor growth and progression in a variety of cancers including mast cell diseases gastrointestinal stromal tumor acute myeloid leukemia Ewing sarcoma and lung cancer. Phosphorylation at tyrosine 721 of c-Kit allows binding and activation of PI3-kinase.
Function:
This is the receptor for stem cell factor (mast cell growth factor). It has a tyrosine-protein kinase activity. Binding of the ligands leads to the autophosphorylation of KIT and its association with substrates such as phosphatidylinositol 3-kinase (Pi3K).

ATP + a [protein]-L-tyrosine = ADP + a [protein]-L-tyrosine phosphate.
