Application Note:
BL: Use at an assay dependent dilution. 20-272-193147 inhibits plasma antyithrombin III activity in chromogenic thrombin assays in the presence of excess heparin and blocks purified IIa inhibition activity and purified Xa inhibition activity in the absence of heparin. Dot: Use at a concentration of 1 - 5 µ g/ml.

Molecular Weight:
53 kDa. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. Myeloma: NS1 Antithrombin III (ATH III) is a plasma protein synthesised in the liver. Normal plasma levels are 115-160mg/L. ATH III consists of a single polypeptide chain and has a molecular weight of 58kDa. ATH III is a serine protease inhibitor and regulates coagulation by the formation of covalently-liked complexes. ATH III also has a heparin binding site and in the presence of heparin the inhibition of thrombin factors IXa Xa and XIa is enhanced 1000-fold. Low levels of ATH III (functional or concentration) are associated with deep vein thrombosis and pulmonary embolism. Antithrombin assays are based on immunologic enzymatic or clotting properties.
Function:
Most important serine protease inhibitor in plasma that regulates the blood coagulation cascade. AT-III inhibits thrombin as well as factors IXa Xa and XIa. Its inhibitory activity is greatly enhanced in the presence of heparin.

Disease:
Defects in SERPINC1 are the cause of antithrombin-III deficiency (AT3D) [MIM:107300]. AT3D is an important risk factor for hereditary thrombophilia a hemostatic disorder characterized by a tendency to recurrent thrombosis. AT3D is classified into 4 types. Type I: characterized by a 50% decrease in antigenic and functional levels. Type II: has defects affecting the thrombin-binding domain. Type III: alteration of the heparin-binding domain. Plasma AT-III antigen levels are normal in type II and III. Type IV: consists of miscellaneous group of unclassifiable mutations.
