It inhibits hepatitis C viral replication through acting on the crucial 'membranous web' that facilitates RNA replication. Velpatasvir is a pan-genotypic inhibitor of HCV nonstructural protein 5A (NS5A) with activity against genotype 1 (GT1) to GT6 HCV replicons. Velpatasvir has demonstrated a high barrier to resistance in HCV replicon assays and a pangenotypic antiviral activity in patients with genotypes 1-4 HCV infection. It is being tested in combination with other pangenotypic direct-acting antivirals to obtain a well-tolerated, highly effective treatment option for all HCV genotypes.
