Description: SRI-011381is a novel and potent agonist of the TGF-beta signaling pathway fortreatment of Alzheimer's disease. SRI-011381 reversed or preventedneuronal damage consistently by at least 30% for all neuronal markers.As an example, the loss of dendritic integrity with kainic acid and theprotection with SRI-011381 are shown in FIG. 20. The drug also reducedCD68 expression by more than 20%. Saikosaponin A (SSA) is a triterpenoidsaponin with many pharmacological activities, includinganti-inflammatory and antioxidant effects. The effect of SSA on cardiacremodeling and fibrosis, however, remains unclear. The Smad pathwayactivator SRI-011381 eliminated SSA (30uM)-induced protective effects on fibroblasts. The Wnt pathway activatorWAY-262611 eliminated SSA (1 uM)-induced protective effects onendothelial cells. In summary, this study indicates the potentialapplication of SSA in the treatment of myocardial fibrosis in cardiacfibrosis, with different target effects associated with differentdosages.
