Description
The cytokeratins (CKs) belong to the intermediate filaments, which create aCytoskeleton in almost all eukaryotic cells. In contrast to other intermediate filaments, CKs are made up of a highly complex multigene family of polypeptides with molecular masses ranging from 40 to 68 kDa. CKs are generally held to belong to the most fundamental markers of epithelial differentiation, and until now, 20 distinct CK polypeptides have been revealed in various human epithelia. The CKs can be divided into an acidic type A (class I) and a neutral-basic type B (class II) subfamily. CK 10 is an intermediate-sized, acidic type I CK, with a molecular mass of 56.5 kDa, expressed in all suprabasal cells simultaneously with CK 1, but absent in basal cells. Together, CK 1 and CK 10 represent some of the first markers of epidermal differentiation. CK 10 has been implicated as marker for more differentiated cells in squamous cell carcinomas. In Western blotting of cytoskeletal preparations from human epidermis, the antibody DE-K10 labels a single 56.5 kDa band corresponding to CK 10. In two-dimensional immunoblotting of cytoskeletal proteins from human skin, the antibody labels a single dot corresponding to CK 10. Cells labeled by the antibody display a cytoplasmic staining pattern.
