Description
Glutathione (GSH) is an important antioxidant molecule that functions as a cofactor for a variety of enzymes and is involved in various biological processes. Total GSH includes reduced GSH and oxidized GSH (GSSG). The concentrations of GSH and GSSG and their molar ratio can be used for indicating the oxidative stress status.
The SensoLyte® Total GSH Quantitation Assay Kit utilizes the widely used Elmann’s reagent and an optimized enzymatic recycling method for the quantification of total glutathione. Glutathione reductase converts oxidized glutathione to reduced glutathione. Reduced GSH reacts with Ellman’s Reagent [5, 5’-dithiobis (2-nitrobenzoic acid)] and generates 2-nitro-5-thiobenzoic acid (TNB) producing a yellow color that can be detected at absorbance 405-415 nm. The intensity of the color produced is proportional to GSH concentration. The kit also provides 5-sulfosalicylic acid for protein removal from biological samples.
