Piccolo is a presynaptic cytomatrix protein localized at the presynaptic side of synaptic junctions. It is a large, multi-domain protein composed of an N-terminal zinc finger (Zn²⁺), several Piccolo-Bassoon homology (PBH) domains, and C-terminal PDZ and C2 domains. Piccolo is commonly found in association with Bassoon, a structurally related scaffolding protein, and both are key components of the cytomatrix at the active zone (CAZ).As a scaffolding protein, Piccolo plays a critical role in organizing molecules involved in the endocytosis and exocytosis of synaptic vesicles. Recent studies have shown that Piccolo can modulate clathrin-mediated endocytosis by interacting with Abp1, an actin- and dynamin-binding protein.Piccolo is highly expressed in the brain, with lower expression observed in the stomach. It is not detected in other tissues such as the adrenal gland, testis, or pancreas.
