EGFR is a glycoprotein belonging to the ErbB receptor family. EGFR is widely distributed on the surface of mammalian epithelial cells, fibroblasts, glial cells, keratinocytes and other cells. The EGFR signaling pathway affects the physiological processes of cell growth, proliferation and differentiation. EGFR is a tyrosine kinase receptor, which is located on the cell membrane surface. After binding with ligand, it dimerizes to activate autophosphorylation of tyrosine residues. These phosphorylated residues are used to recruit aptamer proteins and the binding site of the tyrosine kinase substrate. The activated receptor complex interacts with proteins to activate the MAPK, Akt and JNK pathways, thereby controlling cell proliferation, differentiation, motility and survival, and tissue development. In addition, in most tumors, most of the epithelial growth factor receptor mutations or overexpression causes the enhancement of downstream signal transduction.
