Beta-Arrestin 1 is one of β-Arrestins (ARRBs), the best-known negative regulators of G protein-coupled receptor signaling. β-Arrestins are versatile and multifunctional adapter proteins that regulate diverse cellular functions, including cell growth, apoptosis and immune responses. Overexpression of beta-Arrestin 1 has been found in various cancers, indicating it is a potential therapeutic target for cancer treatment. Recently, expression of beta-Arrestin 1 in saliva has been identified as a candidate circadian biomarker. Erk1/2 constitutively phosphorylates beta-Arrestin 1 at carboxy-terminal Ser412, which promotes cytosolic localization of the scaffold protein. Dephosphorylation is an essential step of beta-Arrestin 1-mediated receptor endocytosis, but it is not required for receptor desensitization.
