Description
Heterogeneous nuclear RNAs (hnRNAs) which include mRNA precursors and mature mRNAs are associated with specific proteins to form heterogenous ribonucleoprotein (hnRNP) complexes. Heterogeneous nuclear ribonucleoprotein L is among the proteins that are stably associated with hnRNP complexes and along with other hnRNP proteins is likely to play a major role in the formation packaging processing and function of mRNA. Heterogeneous nuclear ribonucleoprotein L is present in the nucleoplasm as part of the HNRP complex HNRP proteins have also been identified outside of the nucleoplasm. Exchange of hnRNP for mRNA-binding proteins accompanies transport of mRNA from the nucleus to the cytoplasm. Since HNRP proteins have been shown to shuttle between the nucleus and the cytoplasm it is possible that they also have cytoplasmic functions. Strucutrally HNRPL contains 2 segments of ~80aa each which are weakly related to each other and to the ribonucleoprotein consensus sequence-type RNA-binding domains of other hnRNP and snRNP proteins.

Applications:
Suitable for use in Immunofluorescence, Flow Cytometry, ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry, Immunocytochemistry. Other applications not tested.

Recommended Dilution:
Immunohistochemistry(Paraffin): 1ug/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.
Western Blot: 1:2000. Detects a band of ~64kD (predicted molecular weight: 64kD).
Flow Cytometry: Use 1ug for 106 cells.
Optimal dilutions to be determined by the researcher.

Hybridoma:
Sp2/0 myeloma cells with spleen cells from Balb/c mice.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
