Description
BRCA1-associated protein (UniProt Q99MP8; also known as BRAP2, IMP, Impedes mitogenic signal propagation) is encoded by the Brap gene (Gene ID 72399) in murine species. BRAP is a Ras effector that controls the sensitivity of MAPK signaling by negatively regulating the MAPK scaffold protein KSR1 (kinase suppressor of Ras 1), allowing adaptations and homeostasis in a complex tissue environment. In the developing CNS, mitogenic/morphogenic molecules from location-specific signaling centers form counteracting gradients and simultaneously guide orchestrated cell proliferation, neuronal differentiation, and tissue morphogenesis. Consistent with its role as a MAPK signaling threshold modulator, BRAP2 is shown to interact with both Nde1 and Lis1 to form a complex that provides proper buffer strength to set up the signaling threshold according to the local concentration of mitogens and morphogens. The Nde1-Lis1-BRAP complex helps prevent excessive activation of MAPK under high local mitogenic activity in the midline and promote the self-renewal of progenitors that are programmed to generate a large cohort of neurons, but are localized distant from the midline.

Applications:
Suitable for use in Western Blot and Immunocytochemistry. Other applications not tested.

Recommended Dilution:
Western Blot: 0.5ug/ml detects exogenously overexpressed mouse BRAP in 25-50ug of lysate from transfected HEK293T cells
Immunocytochemistry: 5ug/ml detects EGFP-tagged mouse BRAP in transfected COS-7 cells
Note: DO NOT HEAT SAMPLES PRIOR TO GEL LOADING. BRAP protein tends to form insoluble aggregates upon heating.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
