Recognizes human Bcl 2. Shows consistent negative reactions on reactive germinal centers and positive staining of neoplastic follicles in follicular lymphoma.
Description
B0807-03M is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. This antibody may also be used in distinguishing between those follicular lymphomas that express Bcl 2 protein and the small number in which the neoplastic cells are Bcl 2 negative. Anti-Bcl 2 has been used as a predictive biomarker for recurrence in the case of radical prostatectomy for prostate cancer, and has shown promise as a prognostic marker in the case of cancer of the breast and non-small cell carcinoma of the lung.

Applications:
Suitable for use in Immunohistochemistry. Other applications not tested.

Recommended Dilution:
Immunohistochemistry (Paraffin, frozen): 1:5-1:25. Cytoplasmic staining pattern.
Optimal dilutions to be determined by the researcher.

Immunohistochemistry Preparation and Pretreatment:
1. Cut 3-4um sections of formalin-fixed paraffin embedded tissue and place on positively charged slides; dry overnight at 58ºC.
2. Deparaffinize, rehydrate and epitope retrieve; the preferred method is the use of Heat Induced Epitope Retrieval (HIER) techniques in conjugation with a pressure cooker. The preferred method allows for stimultaneous deparaffinization, rehydration and epitope retrieval. Upon completion, rinse with 5 changes of ddH2O.
3. If using a HRP detection system, place slides in peroxide block for 10 minutes; rinse.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
