Specificity
Reacts with the ca.55kD c-Fos protein. Additional and unidentified bands at ca.65 and ca.75kD were also observed in Western blots of EGF-stimulated A431 and T47D cell lysates. Reactivity has been confirmed in Western blotting with human U2-OS, SW480, T47D, EGF-stimulated A431, NCI-H69, and jurkat cell lysates. The 55kD target band was also identified in mouse NIH-3T3 and rat PC-12 cell lysates. Based on amino acid sequence homology, reactivity is also expected with porcine, bovine, cat, Chinese hamster, and golden hamster c-Fos proteins.
Description
The c-Fos oncoprotein is a member of the AP-1 family of transcription factors, which includes other Fos-related (FosB, Fra-1, Fra-2) and Jun-related (c-Jun, Jun B, Jun D) proteins. The downstream effects of c-Fos induction are numerous and in addition to roles in proliferation, transformation and apoptosis, c-Fos has been implicated in the differentiation of several cell
types. c-Fos also plays a role in the regulation of gene expression during proliferation and in the responses of postmitotic neurons to trans-synaptic stimulation. c-Fos is also required for the development of a malignant tumor. c-Fos is localized in the nucleus of normal human tissues.

Applications:
Suitable for use in Western Blot. Other applications have not been tested.

Recommended Dilution:
Western Blot: 1-3ug/ml
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
