Specificity
Recognizes a highly conserved epitope of the human CD3 molecule expressed by T lymphocytes. Species Crossreactivity: bovine, canine, equine, Rhesus Monkey, porcine, chicken, mouse, duck, koala, harbor porpoise, alpaca, Cynomolgus monkey, spotted hyena, sea lion, feline, Amazon parrot, raccoon, Great horned owl (Bubo virginianus), bullfrog, Xenopus and rabbit.
Description
The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. In cortical thymocytes, CD3 is predominantly intracytoplasmic; however, in medullary thymocytes, it appears on the T cell surface. CD3 antigen is a highly specific marker for T cells and is present in majority of T cell neoplasms. CD3 antigen is expressed on approximately 60–80% of human peripheral blood lymphocytes, 20–40% of splenic lymphocytes, the majority of T-CLL and approximately 70% pf T-All.
The CD3 complex consists of five different polypeptide chains (designated as gamma, delta, epsilon, zeta, and eta) with MW ranging from 16-28kD.

Applications:
Suitable for use in Flow Cytometry, Immunohistochemistry, Immunofluorescence and Western Blot. Other applications not tested.

Recommended Dilutions:
Flow Cytometry: 1:50-1:100 Requires membrane permeabilization
Immunohistochemistry (Frozen): 1:100
Immunohistochemistry (Paraffin): 1:100. Requires antigen retrieval using heat treatment using Tris/EDTA buffer pH 9.0 prior to staining of paraffin sections.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
