Cytoskeletal Desmin extract of chicken gizzard
Specificity
Recognizes chicken Desmin, which is expressed in smooth and striated muscle cells and their tumors, i.e. rhabdomyosarcoma and leiomyosarcoma. Species Crossreactivity: Human, mouse, rat, hamster, canine, porcine, rabbit.
Description
Desmin (53kD) exhibits a high degree of tissue specificity, its expression being predominantly confined to all types of muscle cells (cardiac, skeletal and smooth muscle). Regulation of Desmin expression is stage and tissue-specific, since it is induced during terminal development of, for example, skeletal muscle cell differentiation. In skeletal and cardiac muscle cells Desmin is localized in the Z-disk region and at the intercalated disk. The expression pattern of Desmin in smooth muscle is much more heterogenous.
Coexpression of Vimentin and Desmin has been observed in tumors derived from muscle tissue, i.e. rhabdomyosarcomas and leiomyosarcomas. Furthermore, during myocard disfunction dramatic changes in the distribution of Desmin have been observed.

Applications:
Suitable for use in Immunohistochemistry, Western Blot and Immunocytochemistry. Other applications not tested.

Recommended Dilutions:
Immunohistochemistry (Frozen): 1:100-1:200 using ABC method.
Western Blot: 1:100-1:1000
Optimum dilutions to be determined by researcher.

Hybridoma:
Sp2/0-Ag14 myeloma cells with spleen cells from a mouse immunized with a cytoskeletal Desmin extract of chicken gizzard.

Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for 12 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
