Description
Granzyme A, also known as CTLA-3, is the most abundant serine protease found specifically in the cytotoxic granules of cytotoxic T lymphocytes and NK cells. It is released during cellular activation and enters target cells through perforin channels. Granzyme A mediates target cell killing inducing caspase-independent cell death. Human granzyme A is synthesized as a precursor with a signal peptide, a two amino acid propeptide, and a mature chain. Mature human granzyme A shares 71% amino acid sequence identity with mouse granzyme A.

Applications:
Suitable for use in Immunoprecipitation, Immunocytochemistry, Flow Cytometry and ELISA. Other applications not tested.

Recommended Dilutions:
Immunoprecipitation: 25ug/ml using conditioned cell culture medium spiked with recombinant human Granzyme A.
Immunocytochemistry: 8-25ug/ml using immersion fixed human peripheral mononuclear cells (PBMCs)
Flow Cytometry (Intracellular Staining): 2.5ug/10e6 cells using NK-92 human natural killer lymphoma cell line fixed with paraformaldehyde and permeabilized with saponin
Optimal dilutions to be determined by the researcher.

Storage and Stability:
Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
