Immunogen
Synthetic peptide ccorresponding to the sequence (...DFme2KTD...), where me2K corresponds to dimethyl-lysine at position 79 of histone H3 conjugated to KLH. The immunizing sequence is identical in human, mouse, rat and yeast.
Specificity
Recognizes human dimethyl-histone H3 (Lys79) at ca.17kD.
Broad species crossreactivity is expected based on sequence conservation.
Description
Histone H3 is one of the five main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N-terminal tail, H3 is involved with the structure of the nucleosomes of the 'beads on a string' structure.

Applications:
Suitable for use in Western Blot, Dot Blot and Histone-Peptide assay. Chromatin Immunoprecipitation (ChIP) has been reported by an independent laboratory. Other applications not tested.

Recommended Dilution:
Western Blot: 1:4000 Recombinant Histone H3 or acid extracted HeLa cell proteins were resolved by electrophoresis, transferred to nitrocellulose and probed with H5110-14R1. Proteins were visualized using an IgG (HRP) goat anti-rabbit and a chemiluminescence detection system.
Peptide Dot Blot Analysis: 1:2500 dilution of this lot detected the immunizing peptide containing dimethyl-lysine 79. Some crossreactivity with peptide containing monomethyl-lysine 79 was also seen. No crossreactivity to other related histone peptides was found. The antibody did not recognize a histone peptide containing the dual modification of dimethyl-lysine 79 and phospho-threonine 80.
Histone-Peptide Specificity Analysis: 1:25-1:1600 dilution of this lot were incubated with histone H3 peptides containing various modifications conjugated to Luminex® microspheres. Peptide containing dimethyl-lysine 79 was readily detected. Some crossreactivity with peptide containing monomethyl-lysine 79 was seen. The antibody did not detect a histone H3 peptide containing the dual modification of dimethyl-lysine 79 and phosphothreonine 80.
Optimal dilutions to be determined by the researcher.

Storage and Stability:
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
